Effect of reactive inflammation in osteoarthritis on extracellular traps formation by circulating neutrophils

The results of recent studies demonstrating the presence pro-inflammatory mediators in osteoarthritis (OA), as well known similarity histological signs inflammation rheumatoid arthritis (RA) and OA, suggest an important role NETosis immune OA patients. Our objective was to assess ability blood neutrophils from ОA patients generate spontaneously after vitro induction, impact reactive synovitis on dynamics indexes. Thirty-one with verified 30 healthy volunteers were included into study. Circulating isolated one-step density gradient centrifugation using double layers Iohexol gradient. NETs induced by phorbol-12-myristate13-acetate (PMA). These parameters comparable study groups. Mean percentage spontaneous significantly increased comparing controls. Usage PMA, inducing agent accompanied significant increase form NETs. Transition characterized further PMA-induced acute stage disease are higher than state. growth rate NET is 3.74 times upon exacerbation. Statistically peripheral extracellular traps found, depending osteoarthritis. data obtained opportunity for participation circulating via pathogenesis OA.
